موارد مصرف

اين واكسن براي ايجاد ايمني فعال بر عليه عفونتهاي NeisseriaMeningitidis (مننژيت ،سپتي سمي) در افراد با سن بيش از 2 سال كه در نواحي اپيدميك و آندميك زندگي مي كنند،افرادي كه با بيماران مبتلا به بيماري مننگوككي تماس دارند (همراه پيشگيري مناسب با آنتي بيوتيك ها); كاركنان پزشكي و آزمايشگاهي كه احتمال تماس با عامل بيماري زا براي آنها وجود دارد،مسافريني كه قصد عزيمت به نواحي آلوده را دارند;بيماران داراي نقص در جزء انتهايي كمپلمان و بيماران فاقد طحال فعال مصرف مي شود.

موارد منع مصرف

در صورت بارداري ،ابتلا به بيماريهاي حاد و حساسيت به هريك از اجزاء واكسن نبايد مصرف شود.

عوارض جانبی دارو

واكنشهاي سيستميك شامل سردرد،ناخوشي ،تب ،لرز،آدنوپاتي زير بغل مي باشند و درد،قرمزي و تورم در ناحيه تزريق واكنشهاي موضعي نسبت به اين واكسن هستند.

در صورت بارداري ،ابتلا به بيماريهاي حاد و حساسيت به هريك از اجزاء واكسن نبايد مصرف شود.

فارماكوكینتیك

تزريق واكسن تركيبي باعث افزايش پادتن هاي ضدمننگوككي بميزان 4 برابر در 95% افراد مي شود. مصونيت 10-14 روز بعد از واكسيناسيون ايجاد مي شود و براي مدت 3 سال باقي مي ماند،اگرچه سطوح پادتن هاي مربوطه در طي اين مدت بطور قابل ملاحظه اي كاهش مي يابد.

نکات

1- تزريق واكسن ممكن است در همه افراد مصونيت ايجاد نكند.


2- واكسن را بايد دردماي 2-8 درجه سانتيگراد و بدور از نور نگهداري كرد.فقط از محلول همراه واكسن براي رقيق كردن بايد استفاده كرد و آنقدر تكان داده شود تا پودر واكسن كاملا حل شود. درصورت عدم مصرف بعد از 5 روز بايد آن را دور ريخت.

Meningococcal Polysaccharide Vaccine, Groups A, C, Y and W-135 Combined

DRUG DESCRIPTION

Menomune®- A/C/Y/W-135, Meningococcal Polysaccharide Vaccine, Groups A, C, Y and W-135 Combined, for subcutaneous use, is a freeze-dried preparation of the group-specific polysaccharide antigens from Neisseria meningitidis, Group A, Group C, Group Y and Group W-135. N meningitidis are cultivated with Mueller Hinton agar1 and Watson Scherp2 media. The purified polysaccharide is extracted from the Neisseria meningitidis cells and separated from the media by procedures which include centrifugation, detergent precipitation, alcohol precipitation, solvent or organic extraction and diafiltration. No preservative is added during manufacture.

The 0.78 mL vial of diluent contains sterile, preservative-free, pyrogen-free distilled water and is used for reconstitution of product supplied in 1 mL vials. The 6 mL vial of diluent contains sterile, pyrogen-free distilled water to which thimerosal (mercury derivative) 1:10,000 is added as a preservative. The 6 mL vial is for reconstitution of product supplied in 10 mL vials. After reconstitution with diluent as indicated on the label, the 0.5 mL dose is formulated to contain 50 µg of “isolated product” from each of Groups A, C, Y and W-135 in an isotonic sodium chloride solution.

Each dose of vaccine is also formulated to contain 2.5 mg to 5 mg of lactose added as a stabilizer.3 The vaccine when reconstituted is a clear colorless liquid.

Potency is evaluated by measuring the molecular size of each polysaccharide component using a column chromatography method as standardized by the US Food and Drug Administration (FDA) and the World Health Organization (WHO)4 for Meningococcal Polysaccharide Vaccine.

THIS VACCINE CONFORMS TO THE WORLD HEALTH ORGANIZATION (WHO) REQUIREMENTS.
